米Sangamo Biosciences社は、第1世代のZinc Finger Nuclease(ZFN)を用いてエイズ患者から取り出した造血幹細胞のCCR5遺伝子を破壊。拡大培養して患者に戻すex vivoのゲノム編集療法の臨床試験を新たにスタートさせるほか、血友病B患者に対して、ZFNを用いて血液凝固第IX因子遺伝子を組み込むSB-FIX、ムコ多糖症(MPS)1型を対象にα-L-イズロニダーゼ(IDUA)遺伝子を組み込むSB-318などin vivoのゲノム編集療法の臨床試験を実施します。in vivoのゲノム療法はいずれも、アデノ随伴ウイルスベクターを使って、ZFNと治療用蛋白質遺伝子を肝臓に送達し、治療用遺伝子はアルブミン遺伝子座に発現させる戦略で、実施されれば世界初のin vivoのゲノム編集療法となります。
